Summary: | 碩士 === 國立中興大學 === 生命科學院碩士在職專班 === 95 === In order to solve antiadhesion after abdominal operation, we utilized chitosan, gelatin, polyvinylalcohol (PVA) or blend of them with different crosslinking agents to make a temporary hinderance between incision and other tissues in the abdomen. First, we tried to fabricate chitosan/PVA or gelatin/PVA membranes with different ratio, and membranes were crosslinked by 1% glutaldehyde. However, the mechanical properties of these membranes became weak as they encountered tissue fluid. Besides, pure chitosan membrane was reduced by base treatment, and then chitosan membrane expressed better mechanical properties. Although pure chitosan membrane still expressed better mechanical properties in tissue fluid, it was not convenient to be utilized for operation because pure chitosan with base treatment showed better rigid but worst plastic. Therefore, we tried to make better plastic chitosan and gelatin gels to hinder incision after abdominal operation. In order to facilitate chitosan or gelatin gelation, we used higher concentration of glutaldehyde for curing. Higher concentration of the crosslinking agent would make gel faster, molecules assemble, and capacity of adsorbance. So we compared with different concentration of chitosan and gelatin gel cured by 2% glutaldehyde. All evaluation were examined by subcutaneous tests, swelling ratio tests, and antiadhesive tests in abdominal cavity of the rabbit. Finally, we found 20% gelatin with 2% glutaldehyde prevented from adhesion better after abdominal operation.
