The tradition assays have the disadvantages of time consuming, complexity in procedure, and high reagent cost. By applying the microfluidic technologies, a fast, simple, and economical assay can be achieved. Ovarian cancer is the most lethal disease of gynecologic field. The most important determinant of prognosis is early diagnosis. One of the diagnosis methods is the quantification of CA-125. It has been commercialized and widely used for its high sensitivity and specificity. In this work, the CA-125 assay was successfully carried out on the microtiter plate provided by the assay kit, the regular microtiter plate, the in-house made microtiter plate and the microfluidic disc. Our experimental results showed that the assay sensitivity was around 10 U/ml on all the microtiter plate mentioned above. For the preliminary results on the microfluidic disc, it also showed linear relationship. In addition to the assay, a microfluidic detection system with the capability of automatic positioning, detection, and programmable spinning control was also established. The success of this study may offer clinical offices a cost-effective diagnostic tool, which will have a significant contribution to human healthcare.
