Summary: | 碩士 === 朝陽科技大學 === 應用化學系碩士班 === 95 === Macrolide antibiotics are a group of antibacterial compounds that display similar antibacterial properties and are active against Gram-positive and some Gram-negative bacteria. Veterinary drugs are used in animal husbandry to cure diseases or prevent sickness. They can also increase growth efficiency and calm stressed animals during transport. However, over use of these drugs may lead to residues in the edible milk, eggs and tissues.
Colloidal Silver (10-20 nm) was applied as a matrix for the analysis of macrolides residues in eggs by laser desorption/ionization mass spectrometry. In contrast with conventional matrix-assisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF MS), the matrix background interference in the low mass range was significantly reduced by using colloidal silver matrix. With good reproduction, the desorption and ionization of the macrloides were successful through the thermally driven process. Calibration curve were prepared for all macrolides and the range were from 1.5×10^2 to 5.0×10^5 μg/Kg and good linearity was achieved over the concentration range (r>0.996). The detection limits of the developed method(S/N=3)for five macrolides were 1.0×10^2 to 20.3×10^3 μg/Kg.
