The prepared materials were treated on NCTC clone 929 (L929) and analyzed by MTT assay to detect the cell survival rate, by DNA fragmentation assay to analysis its gene toxicity and by Western blot assay to detect the signal transduction pathway of treated cells. Results were compared using one way analysis of variance (ANOVA). Differences in treatment means were analyzed using Student-Newman-Keul’s test and were considered statistical significance at P<0.05. Results: All experiment groups showed cytotoxicity when treated with L929 cells when compared with control group. (P<0.05) The cytotoxicity were increased as concentrations and treated time increased. (P<0.05) There do not show any DNA ladder in all experimental grouops. The L929 p53 expression showed time dependent increased in experimental groups. (P<0.05) Conclusion: From in vitro study showed that MTA or CH mixed with GA which existed lower toxicity.
