In this study, the pharmacokinetics of Oleuropein in rabbits were evaluated. A simple high performance liquid chromatographic method involving UV detection was modified for determination of Oleuropein in rabbits plasma. A Merck LiChrospher 100 RP-18e (250×4 mm) column was used as the stationary phase and mobile phase of methanol:water(50:50) and flow rate at 1.0 ml/min, and 7-hydroxycoumarin solution as the internal standard. The UV absorbance monitored 240nm. The pharmacokinetics of Oleuropein in the rabbit after intravenous bolus administration of various doses (20、40、60mg/kg) were well described by two compartment open model. The elimination half-lives were 9.390±3.399、24.986±5.646、610.773±167.927 min;mean volume of distribution were 0.932±0.258、1.854±0.170、24.517±14.658 L;the area under the plasma level-time curve were about 474.516± 46.446、685.864±28.127、1075.446±110.386 μg.min/ml;the mean drug clearance were 0.125±0.009、0.179±0.008、0.174±0.018 L/min.
