Summary: | 碩士 === 長庚大學 === 醫學生物技術研究所 === 95 === Human metapneumovirus (hMPV) is a newly identified virus. It causes severe respiratory disease in children and elderly people. To study the seroprevalence of hMPV in Taiwan, we develops the indirect immuno-fluorescence assays based on nucleocapsid (N) recombinant protein for detection of hMPV IgG antibody in human sera. A total of 1120 human sera from health people, including 516 male and 604 female, were examined by ELISA system based on N recombinant protein. The data show that the positive rate of each age group in Taiwan was 5.3%(5/94) (0-0.6 years), 5.3%(5/94) (0.6-1 years), 39%(39/100) (1-2 years), 43%(43/100) (2-3 years), 45.1%(43/51) (3-4 years), 53.1%(26/49) (4-5 years), 62%(62/100) (5-6 years), 71.1%(69/97) (6-10 years), 86%(86/100) (10-20 years), 84%(110/131) (20-30 years), 90.6%(96/106) (30-50 years), 91.8%(90/98) (>50 years). Human metapneumovirus is circulating in Taiwan and the first infection usually occurs in the age group of 1-2 years. The positive rate of hMPV in male is the same with the female. We also detected 13 paired sera from children with hMPV infected, the data indicated that the 10 of 13 (77%) human had a seroconversion. The ELISA based on N recombinant protein is a useful tool for seroepidemiological study of hMPV and can apply to clinical
diagnosis.
