Stem cells, believed to possess certain characteristics including self-renew, pluripotency, proliferation, longevity and differentiation, can be a valuable source for neuronal transplantation. The use of fetal tissue is controversial due to ethical and religious issues. Bone marrow stem cells have been proved the ability to differentiation in different germinal layers. Bone marrow stem cells exist two kinds of stem cells at least,Including,the hematopoietic and the mesenchymal stem cells.however,the number of bone marrow stromal cells is relatively low in adult.(Rao and Mattson 2001) Alternative sources of stem cells are therefore desirable. We have reported the successful quantities in vitro attempt at directing the transformation to neuron of mesenchymal cells in Wharton`s jelly of human umbilical cord.
Our aim is to find the association gene and protein during the stem cell transformation into neuron. Here we used neural condition medium(NCM) to treat human umbilical cord mesenchymal stem cells(HUMSC) with 0、3、6、9 days then collect their condition medium on 0、3、6、9 day and total RNA. We try to find out the gene and protein changes during the mesenchymal stem cell transformation into neuron with microarray and cytokine array。
At first,we incubate human umbilical mesenchymal stem cell in the different serum concentration and cell density to observe the change of the surface markers of HUMSCs. Our data show that the increase percentage of CD29 expression on incubated in 2%FBS is more than incubated in 10%FBS.above these we find the surface marker change evidently while the HUMSCs incubated between high and low cell density. Therefore,different incubation condition will observe different characteristic of HUMSCs and the percentage may be different during the HUMSCs transformation into neuron.
