Summary:碩士 === 國立陽明大學 === 公共衛生研究所 === 94 === βig-h3, a extracellular martrix (ECM) protein , was induced by TGF-β,regulated cell adhesion and migration .By realtime PCR, we found that βigh3 mRNA was overexpressed in esophageal carcinoma. There was a A/G single nucleotide polymorphism at -870 of promoter region of βig-h3.By luciferase assay we found that A allele had high transcriptional activity than G allele . After sequence 1Kb promoter region, no mutation was found in this 1Kb promoter region of tumor tissue and adjacent normal tissue of twelve patients. While the loss of heterozygosity(LOH) was observed in 5 heterozygous patients. And the expression of βig-h3 mRNA was no difference among AA,AG,GG genotype, suggest that A/G single nucleotide polymorphism was not a major factor to regulated expression of βig-h3 mRNA. Furthermore, we collected 175esophageal carcinoma patients and 175 controls and genotyped by TaqMan assay. There was no association of this A/G single nucleotide polymorphism and the susceptibility of esophageal carcinoma in this study.
