In the process of bone formation, mechanical stretching may induce molecules to degrade extracellular matrix and inhibit the number of osteoclasts. Therefore, we test whether osteoprotegrin and MMP-3 would respond to the stretching stimulation in MG63 osteosarcoma cell line. RNA and protein were collected after MG63 stretched by Flexercell elongation system (2.79-9.75%) for 1, 8, 16 or 24 hours under frequency of 1 Hz or 0.1 Hz. RT-PCR, real-time PCR, Northern blot and Western blot analysis indicated that both OPG and MMP-3 were upregulated by stretching but the folds of increase may vary depending on the frequency and time point of mechanical stimulation.
