Description
Summary:碩士 === 國立高雄師範大學 === 化學系 === 94 === The Ginkgo biloba L.extracts have been proven to prevent of thrombus, Alzheimer's disease, illness of blood vessel of heart, and are popularly used for health in recent years. However, there is no swift, convenient and efficient way to analyze the Ginkgo biloba L. extracts. In this study, using solid phase extraction on-line with HPLC to analyze Ginkgolides A (GA), Ginkgolides B (GB), Ginkgolides C (GC) and bilobalide (BB) in Ginkgo biloba L.extracts were investigated. The investigation included, the choice of chromatography column and mobile-phase, the injection of samples and the condition for absorption and rinse. It was found that the optimum operating condition for HPLC was to use C-18 reverse phase column. The mobile-phase was set to H2O / MeOH / Isopranol = 68/20/12, with the flow rate 1.0 ml/min and the detecting wavelength was set at 219 nm. On the other hand, the optimum condition for on-line solid-phase extraction was that maximum injection is 300 μL. No strong solution for elution was needed. And the samples were washed with 100 μL, 10 % methyl alcohol. By using the optimal conditions, the enrichments compared with direct HPLC analysis of GC, BB, GA, and GB are 14.6 , 14.1 , 13.3 and 14.0 , respectively. And the low limits of detection(LOD)are 0.3 , 0.5 , 0.5 and 1.2 ppm respectively for the method.
