In human and animals, long-term use of glucocorticoids such as dexamethasone (DEX) possibly will lead to metabolic syndrome, including obesity, insulin resistance, glucose intolerance, hyperglycemia and diabetes. Aspirin, a non-steroid anti-inflammatory drug, has a similar anti-inflammation pathway as DEX. Therefore, the aim of this study was to explore whether DEX and aspirin induce metabolic disorder through altering chromium distribution in normal and obese mice. Diet-induced obese (DIO) C57BL/6JNarl mouse model was used to mimic human obesity. The mice were randomly divided into normal diet (n=19) and high-fat diet feeding group (n=18). Tissue chromium and blood glucose levels were assayed after DEX (2 mg/kg) or aspirin (100 mg/kg) injection. Results indicated that high-fat diet feeding could successfully induce hyperglycemia and obesity in C57BL/6JNarl mice (P<0.01). In DIO mice, after a DEX and an aspirin injection, chromium concentration in liver, fat and skeletal muscle were significantly decreased (P<0.01), and were significantly increased in bone (P<0.001). The chromium redistribution was not significant in normal diet group. In conclusion, chromium reduction in liver, muscle and fat tissue in DIO mice may contribute faulty glucose metabolism after a DEX or an aspirin injection. 毛嘉洪 2006 學位論文 ; thesis 78 zh-TW
