Neu5Ac and its derivatives have been used to treat several diseases, including neuropathic diseases, anti-inflammation and anti-tumor applications. In this study, the effects of cultivation agitation speed, aeration rate, pH, induction time, induction concentration and temperature on the production of sialic acid aldolase by E. coli JM109/pnanA in a batch fermenter were investigated. A maximum sialic acid aldolase production of 1480 U/l was attained when agitation speed = 500 rpm, aeration rate = 4 vvm, pH = 6.5, induction time OD600 around 3, NaCl induction concentration = 150 mM and induction temperature = 37卅aC.
