Summary: | 碩士 === 中華醫事學院 === 生物科技研究所 === 93 === The Sweet osmanthus (Osmanthus fragrans) is an unique medicine herb on Eastern, and was recorded for the medicinal purposes in the ancient medical books. Previous researches indicated that the Sweet osmanthus contains many anti-oxidative constituents. It may protect against the allergic asthma injury, and moderate liver and nervous system function in mice. However, the effects of Sweet osmanthus in the mice brains are still unknown. The purpose of this study was to understand the peroxidation status of brain in OVA-treated asthma mice, and the protective effect of Sweet osmanthus was further examined. The BALB/c mice were divided into five groups: NC (negative control) group, OVA treated group (OVA), OVA and low dose Sweet osmanthus treated group (100 mg KFE/kg/BW, OLK), OVA and medium dose Sweet osmanthus treated group (800 mg KFE/kg/BW, OMK), OVA and high dose Sweet osmanthus treated group (1000 mg KFE/kg/BW, OHK). The mice were fed for 28 days in each group. The OVA antigen was given either by abdomen injection or spray inhalation during the feeding period. After 40 days, the mice were sacrificed and brains were collected for following tests. It showed that the monoamine oxidase (MAO) enzyme activity is significantly (P<0.05) increased in OVA group. However, there were no significant differences on MAO-B activity in OLK, OMK, OHK, and NC groups. It also showed that the catalase enzyme activities are significantly (P<0.05) increased in OVA and OLK groups. However, there are no significant differences on catalase enzyme activity in OMK, OHK, and NC groups. The glutathione (GSH) contents of OLK, OMK, and OHK groups are significantly higher than NC and OVA groups. However, there are no any differences among groups in the TBARS and Vit C assay items. In conclusion, the study demonstrated that Sweet osmanthus extract can protect against peroxidative injury in OVA-treated asthma mice brains.
