Description
Summary:碩士 === 國立陽明大學 === 醫學生物技術研究所 === 93 === For molecular diagnosis of the leukemia, both traditional hybridization assay such as FISH and PCR base method require fluorescence or isotope labelling, which is expensive and time-consuming, To meet the growing market needs in the 21st Century, advance devices must link high performance, speed, simplicity and low cost .DNA biosensor is a good candidate for leukemia genetic diagnosis. DNA biosensors draw much attention due to their tremendous promise for obtaining sequence-specific information in a faster, simpler and inexpensive manner compared to traditional hybridization assays. A quartz crystal microbalance (QCM) would be a powerful alternative for sensing the complementary DNA strands. The instrumentation of a QCM system is quite simple, and no prior chemical modification is needed. We developed a tool for leukemia molecular diagnosis based on QCM system. On the surface of quartz crystal electrode, we increase the surface roughness by employing Au nano-particles. By testing the Bcr-Abl complementary sequence, we found that the signal were significantly amplified. The method of Au-NPs still cannot apply to clinical sample. We expect Au-NPs modification of QCM Biosensor can be a new tool for real medical diagnosis device, and offer a new method for Leukemia diagnosis.
