By real-time PCR, we observed overexpression of TGFBI mRNA in nine out of ten esophageal carcinoma tissues and ten esophageal carcinoma cell lines, but not in adjacent normal tissues. We have generated a TGFBI polyclonal antibody, which recognized a 68 Kd TGFBI protein in the cell lysates of esophageal carcinoma cell lines (CE48T/VGH, CE81T/VGH and CE146T/VGH), and a 60 Kd secreted protein in culture medium. The different molecular weight of TGFBI protein in cell lysate and culture medium was probably due to protein processing. By immunohistochemistry, TGFBI was major located in extracellular matrix in esophageal carcinoma tissues, while weak signal was detected in nucleus and cytoplasm. The data revealed that recombinant TGFBI protein could promote adhesion of esophageal carcinoma cell lines CE48T/VGH, CE81T/VGH and CE146T/VGH, and enhance migration of CE81T/VGH cell line. Thus, TGFBI may play an important role in carcinogenesis of esophageal carcinoma. We also developed a competitive enzyme-linked immunosorbent assay to assay TGFBI protein in the sera of esophageal cancer patients. Serum samples were collected from 123 patients with esophageal carcinoma and 47 normal individuals. The TGFBI protein concentration were elevated in sera from esophageal cancer patients (11.4±4.5μg/ml) compared with serum samples from normal individuals (4.9±3.8μg/ml) (p<0.0001). This method provides a useful assay for TGFBI serum protein, a molecule which may be a serum marker helpful in determining esophageal carcinoma tumor status.
