To study the bending rigidity of a single domain on a multiple-domain vesicle, we use differential confocal microscopy with nanometer sensitivity to measure the out-of-plane fluctuations on each domain. Bending rigidity on each domain is calculated from the measured fluctuation amplitudes. We found the bending modulus extraordinarily small right on the boundary.
