Lipid mixtures composed of particular proportion of DOPC, sphingomyelin and cholesterol were used. To study the bending rigidity of a single domain on a multiple-domain vesicle, we use differential confocal microscopy with nanometer sensitivity to measure the out-of-plane fluctuations on each domain. Bending rigidity on each domain is calculated from the measured fluctuation amplitudes.
We found the bending modulus extraordinarily small right on the boundary. This explains the extremely small radius of curvature required in the budding process.
