Description
Summary:碩士 === 國立屏東科技大學 === 獸醫學系 === 93 === Goose parvovirus ( GPV ) is a highly fatal disease in causing fibrinonecrotic enteritis, perihepatitis and pericarditis of goslings and muscovy ducklings. The recovered geese are suffering from intestinal- malasorptional stunting and serositis. Fourteen old and one new Taiwan isolates of GPV was propagated in the allantoic cavity of 11- to 14-day-old embryonated goose eggs. The collected allantoic fluid oft the goose embryos with petechial hemorrhage were sequenced of their nucleotides and duduced amino acid of GPV-VP2 in comparison with the database in international genbank . The in difference nucleotide among the 15 GPV isolates ranged from 0.0% to 5.4%, 0.1% to 5.6%, 2.7% to 5.2% and 3.4% to 6.5% in comparison with other Taiwan, European, and Chinese isolates respectively. Although GPV is belong to DNA virus, the nucleotide variations among GPV isolates is reasonably high. In the immunoprotectivity test, a GPV-VP2 subunit protein expressed in prokaryotic expression system for developing the vaccine, three of uniform fold self-prepared GPV live vaccine, two of commercial GPV inactivated vaccine, and one of commercial attenuated vaccine was immunized in geese. Not specific pathogen free geese to do animal test, although V325/TW03-VP2 exhibited highly protectivity, but not a good protectivity was found in each immunized groups. For develop a GPV vaccine, need specific pathogen free geese to define a trusty immunoprotectivity test.
