The CIAV supernatant was then binary ethylene imide inactivated and prepared as gel or oil-emulsion type inactivated vaccine for 2 times immunization in a group 3-week-old SPF chickens, 20 birds per group, a live CIAV vaccine and control group was also included. Five birds per group were evaluated for hematocrit value ( HV ) at 10 and 21 day post immunization ( DPI ) and 10 birds was evaluated enzyme- linked immunosorbent assay ( ELISA ) at 28 DPI. All the experimental chickens were found with normal HV ( >25% ) except all the control and live vaccine immunized groups ( 3 and 2 in Trial 1 and 2 respectively ). The ELISA titer in Trial 1 was found highest in the gel vaccine immunized group ( 5612 ), then the live ( 5120 ) and OEV ( 1441 ) immunized group. The ELISA titer in Trial 2 was found highest in the OEV vaccine ( 6861 ) due to changing the oil- adjuvant, then the gel type ( 5056 ) and live ( 4274 ) vaccine. The Taiwan field isolated CIAV propagated in MDCC-MSB1 cell by suspension cultivation provide good potential in the preparation of high immunoprotectivity of inactivated CIAV vaccine.
