Summary:碩士 === 國立屏東科技大學 === 水產養殖系 === 93 === The aim of this study is to introduce a GFP (green fluorescent protein , Pcmv-EGFP) gene into the oocytes of Barbus tetrazona by electroporating the transgene construction into the sperm. A total of 120 fertilized eggs have been successfully produced, with 68% fertilized and 88% hatchery rates in average. A significant difference was found in fertilized rate, but not in hatchery rate of the electroporated group, as compared with the control group. Ten hours after fertilization, the expression of GFP was detected in 15 out of the 18 embryos as determined by the PCR analysis. Furthermore, the whole embryos of the electroporated group displayed green fluorescence at 18 hours after fertilization. These result suggest that tranagenic Barbus tetrazona can be produced by sperm-mediated gene transfer.
