Factors affecting analysis such as separation temperature, pH and the injection volume of sample solution were investigated to obtain the optimum conditions. By the optimum conditions, the linear range, reproduction and the detection limit of the analysis method will be measured, then apply the developed method to analyze sulfonamides of commercial milk. The on-line SPE is proposed to be a new pre-treatment technique having advantages that to avoid losing samples and it is fast and easy to use. Experimental results indicated that fourteen sulfonamides are well separated in an acceptable time on the reversed-phase C18 column eluted with the gradient elution of formic acid aqueous solution/acetonitrile at 1.0 ml/min flow-rate and to set the detection wavelength at 270nm. The optimal conditions of the on-line SPE analysis are 800μl of injection volume and the sample solution adjusted to pH 5. Analyzing sulfonamides of three kinds of milk by using the new technique, the recoveries ranging from 79 to 119% of fourteen sulfonamides were determined.
