To identify potential cellular proteins interacting with JEV NS5 protein, yeast two-hybrid assay was used to screen the mouse brain cDNA library. One of the positive clones identified using this assay is Ran binding protein 9 (RanBP9 or RanBPM). Functions of RanBP9 involve the regulation of transportation through nuclear pore. Co-immunoprecipitation assay and cell immuno- fluorescence staining were used to demonstrate the interaction of RanBP9 with NS5.
