Summary:碩士 === 國立東華大學 === 生物技術研究所 === 93 === The non-structural protein 5 (NS5) of Japanese encephalitis virus (JEV) encodes RNA-dependent RNA polymerase (RdRp) that is required for replication of the viral RNA genome. To identify potential cellular proteins interacting with JEV NS5 protein, yeast two-hybrid assay was used to screen the mouse brain cDNA library. One of the positive clones identified using this assay is Ran binding protein 9 (RanBP9 or RanBPM). Functions of RanBP9 involve the regulation of transportation through nuclear pore. Co-immunoprecipitation assay and cell immuno- fluorescence staining were used to demonstrate the interaction of RanBP9 with NS5. These results suggest that the interaction of NS5 with RanBP9 may play a role in the function of NS5.
