At first, a cross-linking monomer, (N-Acr-L-Cys-NHBn)2, was synthesized, attached to the surface of the chip and copolymerized with other monomers to form a molecularly imprinted film. During the process of polymerization, ethylene glycol dimethacrylate or N,N’-ethylene bisacrylamide was added as a crosslinker to strengthen the structure of the molecularly imprinted film. We can differentiate traces of bradykinin to the ng/ml scale. However, embedding was still unavoidable. The MIP chips were detected by mass spectrometer to examine the formation of molecularly imprinted film, the effect of removing template and the ability of molecularly imprinted film to recognize the target molecule. During the process of polymerization, a new monomer, 3-acr-5-hydroxy-benzoic acid, was used as a matrix to enhance the intensity of molecular ion. Although the intensity were not consistent with the concentration of matrix in the MIP, it was increased upon adding matrix-monomer. As we expected, no bradykinin was detected by mass spectroscopy after washing. However, the molecular ion was appeared on the spectroscopy in the presence of matrix. Therefore, laser desorption might be an applicable tool to detect and remove the embedded template.
