Nowadays people are still threatened by the microbacterial infections, especially tuberculosis bacterium. However, the infection rate of tuberculosis is higher in Taiwan than in U.S., Europe and Japan. It is necessary to evaluate and reconstruct the screen systems to tuberculosis bacterium in public hygiene and medical treatment. A standard procedure has been developed maturity to separate the bacterium in laboratory operation, but the storage period of specimens resulted in inconsistent inoculation and the evaluations of bio-chip and antibody detection. In the present study, the amount of bacteria in sputum and the storage periods of specimen have been showed to influence the accuracy of inoculation. The tuberculosis bacterium was identified precisely by the bio-chip detection with either liquid culture or the direct sputum inoculation, but false positive results were obtained in the direct sputum inoculation. To avoid the influences of impurity in sputum, 50% sucrose gradient procedure was applied to eliminate the false signals in bio-chip detection. In the other hand, the results from serum antibody determination exhibited a high sensitivity but low specificity in tuberculosis bacterium detection. In conclusions, the combination of sucrose gradient procedure with bio-chip detection conducted a high specific screening approach to detect the tuberculosis bacterium in sputum samples.
