KCS is a disorder of the tear film due to impairment of tear film quantity and /or quality which causes damage to the cornea and conjunctiva, even reduced vision. To date, there is little study of dog tears, particularly in relation to disease states and alteration of tear proteins. From the result of protein analysis by gel electrophoresis using normal and KCS dog tears, a band of ~ 75 kD was found and it showed significant expression between normal and KCS dogs. Sequence data of this band from mass spectrometry indicated that this protein is canine serum albumin. All tear samples were collected from 54 patients and volunteers of Chung-Hsing University teaching veterinary hospital and three local hospitals by Schirmer tear strips. All patients was divided into four groups according to clinical signs, Schirmer tear test results and treatment by cyclosporine. The concentrations of albumin of volunteers and patients were analyzed with software, and results showed that the both tear albumin and total protein of KCS group were much higher than those of other three groups. We found that the albumin in canine serum of KCS group or normal group was in the normal range. In the early stage of KCS, the Schirmer tear test result is not only elevated but also difficult to distinguish with other ocular disease. Our study suggested the alteration of albumin concentration can be a tool of diagnosis in the KCS early stage. With ROC curve, we chose a cut point. When the albumin concentration is over the cut point, this dog has a highly opportunities in the KCS early stage. Similarly, we found the concentration of total tear protein and the ratio of albumin in total tear protein can also be a diagnosis or prediction tool of KCS.
