Solid state fermentation was carried out in specially designed plastic bags(16×8×13 cm) containing 50 g peat moss, agricultural wastes (soybean powder, sugar cane bagasse and rice bran), surfactant (Tween 80) with different moisture content. Substrate was inoculated with 5 mL conidial suspension of T. virens (1×108 conidia/mL), and bags were incubated at 27 ℃ for 7 days then dry for 2 days. The optimal medium of solid-state fermentation consisted of 4 % soybean powder, 0.62 % Tween 80, 1.3 % sugar cane bagasse, 5.3 % rice bran and with 80.7 % of moisture content. Under optimized conditions, the conidial production was 1.1×109 conidia/g-substrate dry weight. However, results showed that, based on central composite design, a medium comprised 5.1 % soybean powder, 0.24 % Tween 80, 1.8 % sugar cane bagasse, 7 % rice bran with moisture content of 86 % was proposed, which gave a yield of 2.4×109 conidia/g-substrate dry weight. To solve the discrepancy, more studies on the search of optimal medium applying central composite design experiment is urgently needed. 謝建元 2005 學位論文 ; thesis 0 zh-TW
