Summary: | 碩士 === 朝陽科技大學 === 應用化學系碩士班 === 93 === In this searsch, the more aqueous stable material of L-ascobic acid derivative, being Magnesium Ascobyl Phosphate( MAP ) was formulate in the hydrogel mask for the stability study. Further more, the addition of Lauric acid in the formulation was pwnwtration enhances whether it world taciliate the MAP release through transdermal penetration. The High-performance liquid chromatography( HPLC ) was capitalized for the analytical work.
From the HPLC results, if was shown that MAP stability was dependent on pH of the formulation. As pH of 7, the hydrogel still residue 90% MAP after 150-day storage. In the transdermal penetration test, the formulation comprising lauric acid provided 30% more MAP release than those without lauric acid when the stomach skin of rabbit was used as a membrane for testing.
As for HPLC analysis, an ODS hypersil column (250 x 4.6 mm ; 5μm ) was used with a mobil phase consisting of 0.5mM Tetrabutylammonium bromide ( TBA-Br ) and 50 mM disodium hydrogen phosphate as a buffer. An UV detector set as 259nm wave length and the elution rate of 1.0mL/min were utilized. In general, each run about eight minutes of time.
