Because of the nephrotoxicity and ototoxicity of Kanamycin A, it is necessary to develop a rapid and sensitive method to determine the concentration in biological fluids. A capillary electrophoresis with laser-induced fluorescence detection method for the determination of kanamycin A has been developed. 6-Carboxyfluorescein succinimidyl ester (CFSE) was used for precolumn derivatization of the non-fluorescent drug. Linearity over three orders of magnitudes was generally obtained. The limit of detection for kanamycin A in aqueous solution was calculated to be 9.1 nM. The relative standard deviation of migration times and peak areas were 0.9% and 9.2% respectively. Combining with a simple cleanup procedure, this method was successfully applied to the determination of kanamycin A in human plasma. The ratio of peak area of kanamycin A / baclofen versus kanamycin’s A molar concentration plot showed a good linearity wite a correlation coefficient (r) of 0.998, and the limit of detection of kanamycin A in plasma was about 3.9 nM.
