Therefore, it is suggested antioxidants may be useful for the prevention and treatment of neurodegenerative diseases. Ligustri Lucidi Fructus is the dry ripe fruits of Ligustrum lucidum Ait. (Oleaceae). In this study the antioxidant activities, and antioxidant constituents of Ligustri Lucidi Fructus, Ecliptae Herba, and Erzhi Wan, were evaluated. The neuroprotecitvie effects of Ligustri Lucidi Fructus on hydrogen peroxide induced oxidative damage in SH-SY5Y cells were examined. The experimental results demonstrated that: 1, Erzhi Wan exhibits stronger antioxidant activity than Ligustri Lucidi Fructus and Ecliptae Herba. Crude extract of Ligustri Lucidi Fructus scavenges superoxide anion, hydroxyl and peroxyl radical, and inhibits lipid peroxidation. Among all subfractions, ethyl acetate fraction possesses highest activities in antioxidation and scavenging superoxide anion; however, chloroform fraction possesses highest activities in scavenging hydroxyl and peroxyl radical. 2, crude extract of Ligustri Lucidi Fructus elevates cell viability, decreases intracellular oxidative stress and reverses the decrease in GSH contents and GSH/GSSG ratio in hydrogen peroxide treated SH-SY5Y cells. Ethyl acetate fraction possesses better neuroprotective activity than chloroform fractions in hydrogen peroxide induced neuronal oxidative damage. Ethyl acetate fraction also decreases intracellular oxidative stress, and reverses the decrease in GSH contents and caspase-3 activation caused by hydrogen peroxide. Thus, Ligustri Lucidi Fructus possesses neuroprotective effects and its active constituents may exist in ethyl acetate fraction. The neuroprotective effects of Ligustri Lucidi Fructus might be due to its phenolic antioxidant activities, and decreasing intracellular oxidative stress, GSH consumption and caspase-3 activation by hydrogen peroxide.
