The goal of this study is to find the better antibody coating method and to standardize the coating protocol. We hope to by using latex beads to amplify the detection signal. We expected to detect antigen at concentration as low as 1 ng/ml. Finally, we hope to apply QCM chip in clinical test, and to supply a unexpensive and quick test. In this research, we use glutaraldehyde plus PEI method and EDC plus PEI method to coate antibody on the chip. After camparing both detection ability, we found EDC plus PEI method was better for coating because it can detect antigen at concentration of 1μg/ml. At the same time, we optimized the concentration of EDC and NHS, and the blocking protocols of glycine and BSA. Finally, we use Latex beads to enhanced the detection signals. Using latex beads, the chip can detect as low as 1ng/ml antigen. In summary, we used EDC plus PEI method is a better way to coat antibody on the chip.
