Influenza rapidly spreads around the world in seasonal epidemics. Because the virus mutates so frequently that created many new virus strains each year, novel strategies for the treatment or prevention of influenza infection are needed and attractive. To date, two principal options to control influenza are vaccines and antiviral drugs. However both of them have certain limitations, such as mutant viruses or drug-resistant viral strains. So, there must be other ways to against influenza infection. In recent years, investigation of traditional Chinese herbs has attracted increasing interest. Some combinations of Chinese herbs have been reported to be effective against respiratory tract infection. Our lab found that Da-Qing-Long-Tang (DQLT) was able to reduced the plaque formation induced by both influenza A/WSN/33 and influenza B/Beijing/184/93-like. However, Yin-Chyau-San (YCS), another traditional Chinese medicine that is frequently prescribed, was effective to inhibit influenza virus. So, we proposed that traditional Chinese herbs might have effective anti-influenza virus activities by increasing host immunity. Therefore, we focus on the effect of traditional Chinese medicine on influenza A virus-induced immune responses. In the present study, we demonstrate in vitro assay for antiviral activity. We found that DQLT had more significant inhibitory effects than YCS on plaque formation induced by influenza A virus. Then, the monocytes isolated from PBMCs were cultured with GM-CSF and IL-4 in complete RPMI-1640 medium were differentiated into dendritic cells (DCs). When cultured with M-CSF in complete RPMI-1640 medium, the monocytes were differentiated into macrophages (Ms). Flow cytometry and ELISA data showed that both DQLT and YCS had no effects on the activity of DCs, but suppressed the activity of Ms. CD3+ T cells were enrichmented from PBMCs using Nylon wool column, and were stimulated with anti-CD3, anti-CD28 plus DQLT or YCS. Results indicated that traditional Chinese medicine induced autologous T cells activation and IFN-γ production. When traditional Chinese medicine were tested on the in vitro DCs (MΦ)/ T cell influenza virus infection models.
