Mac-2 binding protein (Mac-2 BP), one of the identified proteins, is a ~90 kDa secreted glycoprotein that may play roles in cell-cell and cell-extracellular matrix adhesion. It can bind galectin-3 (formerly Mac-2), galectin-1, galectin-7, β-integrins, collagens and fibronectin, respectively. Previous studies have shown that high serum levels of Mac-2 BP correlated with high metastasis and poor prognosis in several neoplasms. It is interesting to observe that Mac-2 BP could be detected in culture media of certain cancer cell lines with high metastatic ability. Mac-2 BP expression (Mac-2 BP staining positive: ≥50% of the neoplastic cells) was observed immunohistochemically in 50% (119/273) colorectal carcinoma. Futher analysis showed that a high expression of Mac-2 BP correlates with mucinous adenocarcinoma (p=0.0084). However, there was no significant difference for the overall survival rates between patients with high and low level of Mac-2 BP expression. To understand the functional role of Mac-2 BP in tumor cells, siRNA was used to inhibit Mac-2 BP expression in NPC-TW02 cell line. The Mac-2 BP mRNA and protein levels were significantly decreased in NPC cells transfected with Mac-2 BP siRNA. Inhibition of Mac-2 BP did not affect the proliferation of NPC cells. On the other hand, the migration of NPC cells treated with Mac-2 BP siRNA was slower than that of control NPC cells in wound healing assays.
