Summary: | 碩士 === 國立陽明大學 === 傳統醫藥學研究所 === 92 === “Huangqin” (root of Scutellaria baicalensis Georgi) is a Chinese medicinal herb. It is generally used for fever, cough, pneumonia, jaundice, hepatitis, and hypertension. Previous investigations have shown that water extract of Scutellaria baicalensis Georgi inhibited the secretion of HBsAg and HBeAg on human hepatocellular carcinoma HepG2/A2 cell line. The aim of our study is to isolate anti-hepatitis B virus compounds. By using human hepatocellular carcinoma cell line, HepG2/A2 as in vitro model, cellular toxicity and HBsAg and HBeAg secretion were measured by MTT and ELISA assays, respectively. The extracts of Scutellaria baicalensis Georgi were further purified by column chromatography. Five compounds baicalein(1), neobaicalein(2), wogonin(3), 2’,5,6’,7-tetrahydroxyflavanone(4)and oroxylin A(5) were isolated by bioassay directed fractionation. The results showed that neobaicalein(2) significantly(p<0.05)inhibited the expression of HBsAg and HBeAg with a little cytotoxicity. Wogonin exhibited an anti-hepatitis B virus activity against the secretion of HBeAg(EC50 = 17.7 μM)and significantly(p<0.05)inhibited the expression of HBsAg without significant cytotoxicity.
We conclude that the anti-hepatitis B virus activity in crude extracts of Scutellaria baicalensis Georgi are from the active compounds such as neobaicalein and wogonin.
