The interaction between immunosuppression and apoptosis after high intensity exercise is not well understood. The aim of this study was to investigate the apoptosis-related gene expression before and after high intensity exercise and it have been developed to help understand the association between exercise-induced leukocyte apoptosis and the immune system response. Twelve subjects ( VO2max 70.4 ± 4.7 ml/ kg / min ) were participated in this study and performed a 3-day consecutive high intensity exercise ( 85 % VO2max for 30 mins ). The blood samples were collected on the following schedule of three days before exercise ( D1 ), after the 1st day exercise ( D1’ ), after the 2nd day exercise ( D2’ ), before the 3rd day exercise ( D3 ), after the 3rd day exercise ( D3’ ), one day later after the 3rd day exercise ( 24 hr ), and three days later after the 3rd day exercise ( 72 hr ). Leukocytes apoptosis-related gene expression was measured by real time reverse transcriptase polymerase chain reaction ( RT-qPCR ). The results indicated that all leukocyte apoptosis-related gene expressions after short term consecutive high intensity exercise were different to D1. The expression of numerous caspases-3 mRNA and leukocyte apoptosis increased simultaneously. Results showed the same trend between leukocyte BAX/Bcl-2 ratio and its mitochondrial transmembrane potential during the period of training. The trends of caspase-3 and caspase-8 mRNA expression were identical to each other and much higher than caspase-9 after short term high intensity exercise. In conclusion, the all leukocyte apoptosis-related genes mRNA elevated after short term consecutive high intensity exercise. The trigger to death receptor and initiation of mitochondria apoptosis pathways may affect each other and happen in the same time.
