Therefore, its effluent water have low pollution for environment, and it has great potential for application. In this work, the application of experimental design for the biosurfactant production by Acinetobacter calcoaceticus RAG-1 from ethanol as sole carbon source were studied in shaking flasks. Then the effect of the rates of agitation in jar fermentor on the biosurfactant production, were studied in relation to the physical and chemical properties, and biodegradability of the biosurfactant. Finally, the microbial growth kinetics and biosurfactant production kinetics were modeled by using Monod equation and the Luedeking and Piret equation, respectively. The experimental results showed that in the shake flasks the optimal medium compositions were 9.16 g/L ethanol, 8.2 g/L KH2PO4, 23.32 g/L K2HPO4, 5.77 g/L (NH4)2SO4 and 0.354 g/L MgSO4.7H2O and the biosurfactant production of 73.312mg/L was obtained after 80 hours of cultivation. The biosurafctant yield were not changed by the rates of agitation in fermenter over 600-800 rpm. According to the Luedeking and Piret model, the bacterial production of biosurfactant was found to be non-growth associated.
