Only a few bacteria were presented in routine culture examination. Bacterial 16S rDNA were detected from the prostatic specimen of 33 chronic prostatitis/chronic pelvic pain syndrome patients in Taiwan. Total DNA extracted from specimen was amplified by PCR using bacteria specific primer pair, and analyzed by denaturing gradient gel electrophrosis. The major or specific DNA band was sequenced and compared with sequences in GenBank. Bacterial DNA was able to amplified from all the samples of patients. Base on cultivation of bacterium, three (9%) of 33 patients with pelvic pain and/or voiding dysfunction were diagnosed as chronic bacterial prostatitis. While among 30 chronic prostatitis/chronic pelvic pain syndrome patients, seven (23%) of them were diagnosed as chronic bacterial prostatitis using molecular method. Due to the limitation of culture method, bacterium was not detected in these samples. The bacteria species associated with chronic prostatitis/chronic pelvic pain syndrome including Escherichia coli, Flavobacterium sp., Mobiluncu curtisii, Prevotella sp., Pseudomonas sp., Rhodococcus sp., Staphylococcus haemolyticus and one unidentified bacteria. Some bacteria may be too less to culture or not growth on the routine medium. There may also be possible that they were non-culturable species. So we can’t find them by culture method. The application of this study is to find the appropriate media to culture and the antibiotic agent for treatment.
