The previous carrier-substrate for limbal epithelial cell culture are amniotic membrane, fibrin or 3T3 fibroblast feeder layer. All of them are derived from animal or human tissues. A minute chance of an unidentified infectious agent can’t be ruled out, and the problems of acquiring tissue or preparing substrate such as fibroblast layer are not easy. We try to develop one artificial chitosan membrane, which is cheap, easily prepared, highly transparent, biodegradable, and highly biocompatible, to be the carrier for limbal epithelial cells culture. This study compare three different conditions of epithelial cells culture. We found the phenotype of transient amplifying cells were partially preserved when stem cells were seeded over chitosan-membrane with amniotic membrane underneath. In spite the chitosan membrane is suitable as a carrier substrate, how to blend the soluble factors in amniotic membrane with chitosan membrane is our future work.
