The purpose of this study was to study the effects of chitosan on the cellular adhesion and growth. The selected cells were platelets, osteoblast and fibroblasts. In part one, the effect of chitosan on human platelet adhesion, activation and aggregation were examined. In part two, the effect of the chitosan on the attachment and growth of fibroblasts and bony cells were evaluated. The results showed that a dose-dependent increase of human platelet adhesion by chitosan (for 15 minutes incubation) was noticed at the range of 0.01 to 1 μg/ml in concentration. Similar results of the platelets’ activation and adhesion were also confirmed by scanning electron microscopy. An increase in numbers of CD61-expression platelets was observed once the chitosan concentrations increased; however, the expression seemed to be reached in maximum if the dose was 200μg /ml. The releasing of PDGF-AB and EGF from platelets was significant increased after incubation with chitosan by a dose-dependent manner. In part two, the growth of primary culture dental alveolar bone cells was increased after chitosan added; however, the growth of human gingival fibroblasts cell line, HGF-1, was not. In addition, the greater bone cell growth was observed in chitosan with 95% deacetylation than that with lower degree of deacetylation. 江孟燦 傅鍔 2004 學位論文 ; thesis 78 zh-TW |
