Most of antimicrobial peptides contain positive charge and low molecular weight, which have the activity against Gram-positive and Gram-negative bacteria, fungi, viruses, and parasites. We had constructed a plasmid containing 6 fragments of antimicrobial peptide gene, pET28KM132-6F. pET28KM132-6F had been transformed into E. coli Rosetta cell. The recombinant antimicrobial peptide KMTrx-6F had been induced by IPTG or lactose, purified by Mono-S and Superdex-75 column, and analyzed by radial diffusion assay. The molecular weight of recombinant antimicrobial peptide KMTrx-6F is about 28 kDa. The minimum inhibitory concentration of recombinant antimicrobial peptide KMTrx-6F is 0.5 μg/ml. The recombinant antimicrobial peptide KMTrx-6F is heat-stable. The antimicrobial activity of recombinant antimicrobial peptide KMTrx-6F is increased when it is dissolved in 0.1% acetic acid. The recombinant antimicrobial peptide KMTrx-6F has the antibacterial ability against Gram-negative aquatic pathogenic bacteria.
