Summary:碩士 === 國立清華大學 === 生物科技研究所 === 92 === Japanese encephalitis virus (JEV) is found throughout Asia from India in the west to Japan in the east. JEV envelope protein plays an important role in cell receptor binding, immunological response, virion assembly and fusion activity at low pH state. After we determining the epitope of JEV envelope protein Domain III binding with monoclonal antibody E3.3 by the chemical shift of NMR HSQC experiment. We used four site- directed mutant Domain III to confirm our result of HSQC experiment. We also used circular dichroism to study the stability of JEV envelope protein Domain III. The △G of Domain III is 5.15 kcal/mole. A recently study(11) shows that JEV envelope protein 261-402 can bind to Heparin, but only 292-402 can’t bind to Heparin. We already titrated Heparin to JEV envelope protein Domain III (292-402). By the HSQC experiment, there is no significant chemical shift in Heparin titrated to JEV envelope protein 292-402. This result is the same with the recently study(11). Now we use JEV envelope protein 261-420 to titrate with Heparin. Try to figure out the binding site of the JEV envelope protein by NMR. This part is still working.
