Tropican Lady and Phal. Tinny Ruby × Doritaenopsis Minho Princess cultured in the dark were used as explants to induce PLBs and subsequent plant regeneration. Plant growth regulators, sugars and genotype were the main factors affecting PLBs formation. Maltose was most effective for stimulation of PLBs formation of Phal. aphrodite Rchb. f. and Phal. Tropican Lady than other sugars. Mannitol inhibited PLBs proliferation. TDZ promoted PLBs formation and subsequent proliferation of Phal. aphrodite Rchb. f. and Phal. Tropican Lady. All tested sugars at 0.03M was the most optimal concentration for PLBs proliferation. TDZ combined with either BA or NAA were more effective for PLBs formation than TDZ alone. 2,4-D 4mg/l and BA 1mg/l combination was more suitable for PLBs formation and proliferation of Phal. Tinny Ruby × Dtps. Minho Princess. Histological sections indicated that PLBs were induced from epidermal or sub-epidermal cells. After transferring PLBs onto regeneration medium and grown in the light normal plantlets were obtained. The combination of 0.03M sucrose, 1mg/l TDZ and 0.5mg/l BA were effective to increase active cell division. Therefore, the PLBs formation was promoted. The DNA content of Phal.
