Summary: | 碩士 === 國立東華大學 === 生物技術研究所 === 92 === According to previous reports, the sulfated polysaccharides and oligosaccharides can inhibit angiogenesis. Evidences show that angiogenesis is regulated to balance by the interaction between stimulators and inhibitors such as growth factors, cytokines, and proteolytic enzymes. In some diseases like cancer, rheumatoid arthritis, and diabetic retinopathy, angiogenesis is out of control in balance condition and causes abnormal angiogenesis. This thesis is to study the inhibition of angiogenesis by sulfated fructooligosaccharides(FOS). First, we used Aspergillus japonicus mycelia to produce FOS and obtained higher purity of FOS by charcoal activated column and ethanol purification process, then carried sulfation processes by replacing the —OH groups in FOS molecules to —OSO3 group. Finally, we examined the sulfated FOS by chick chorioallantoic membrane(CAM) assay to see whether it had anti- angiogenesis effectiveness or not.
Experiment results showed that the conversion of sugars to FOS by Aspergillus japonicus mycelia could over 50%; the purity of FOS by charcoal activated column purification could reach 99%. After FOS sulfation, its color changed from transparent to yellowish-brown. If the further sulfated process ongoing, the color would become dark brown. The result of HPLC analysis showed that the nature of FOS after sulfation was changed. From the result of chick CAM assay, we found that there had obvious inhibition of angiogenesis while the dosage of sulfated FOS was over 1mg. We speculated that sulfated FOS might interact with bFGF and VEGF, which caused the proliferation and migration of endothelial cells, therefore inhibited the angiogenesis.
