Summary:碩士 === 高雄醫學大學 === 醫藥暨應用化學系碩士班 === 92 === The SPIO coated with dextran T-40 was synthesized, and the core size of the iron oxide nanoparticles was 10.32 ± 2.16nm calculated by TEM. The paramagnetic property of SPIO was estimated by SQUID. Dansyl-SPIO and dansyl-CLIO were synthesized by using EDBE to link dansyl chloride and SPIO. Flow-cytometric analysis indicates that the dansyl-SPIO and dansyl-SPIO are specific to the dansyl receptors. The dansyl-CLIO possesses higher ability of labeling to dansyl receptors than that of dansyl-SPIO. Furthermore, the dansyl receptors on B16F1-DNS could be detected by dansyl-CLIO contrast agent for magnetic resonance imaging (MRI). In addition, Herceptin–SPIO was synthesized and used to detect the cells with Her2 gene overexpression. Moreover, Herceptin-SPIO is also proved to be able to monitor the cells with Her2 overexpression by flow-cytometric analysis.
