The wood of Sappan lignum is one of the Chinese Medicines with blood-activating and stasis-eliminating effects. In this study, the effects of Sappan lignum in anti-angiogenesis were examined in human umbilical vein endothelial cells (HUVEC). Our data showed that Vascular endothelial growth factor (VEGF) induced the proliferation of HUVEC in a dose dependent manner. Sappan lignum (25~100μg/ml) significantly inhibited the proliferation of HUVEC induced by VEGF (20 ng/ml). At the concentrations of 25~50 μg/ml, Sappan lignum almost completely blocked VEGF-induced cell proliferation. However, Sappan lignum exhibited overt cytotoxicity at higher doses (> 100 μg/ml). This effect of Sappan lignum is similar to low dose chemotherapeutic agents that pose cytostatic effect and were used as anti-angiogenesis agents. Furthermore, we found that Sappan lignum markedly reduced VEGF (20 ng/ml)-induced migration of HUVEC in a Transwell migration assay. In conclusion, our data suggest that Sappan lignum can markedly inhibit the proliferation and migration-induced by VEGF in HUVEC. Sappan lignum may be an effective herbal drug in anti-angiogenesis.
