In this study, we will use the effective fraction of AF (AFEF) to modulate innate immunity including nautral killer (NK) cell cytotoxic assay, macrophage phagocytosis, splenocyte and lymph node subpopulation differentiate, cell proliferation, cytokine secreting assay and serum immunoglobulin assay. Various doses of AFEF administered to BALB/c mice for 2 and 4 weeks. Primary peritoneal macrophage harvest from mice that administered with various concentration of AFEF was direct to activate phagocytosis. On the other hand, AFEF can significant increase cytolysis effect in NK cell (1500 mg/kg) and that might be involved the activation of T helper cell in activating (CD69). AFEF can also significant increased IFN-production from lymph node cells by ConA stimulation for 48 hours in administration AFEF (1500mg/kg). Further study to understand the mechanism of phagocytosis, in vitro AFEF-treated RAW 264.7 cells were analyzed in this study. AFEF can significant increases RAW 264.7 cell phagocytosis (100, 200 g/ml), by increased nitrite, IL-2 and IFN- secretion.
