One of the proatherogenic properties of oxidized LDL (OxLDL) is its toxicity toward cells including endothelial cells, smooth muscle cells and macrophages. According to the theoretical system of TCM, atherosclerosis is the disease of the blood stagnation. Paeonia suffruticosa Andr. and Carthamus tinctorius L. usually apply to promoting blood circulation to dissipate blood stasis. Therefore , we hypothesized that Paeonia suffruticosa Andr. and Carthamus tinctorius L. can use to treat atherosclerosis. In this study, we investigated the oxLDL-induced HUVECs apoptosis protect by Paeonia suffruticosa Andr. and Carthamus tinctorius L. First, the cytotoxicity of the two drugs in HUVECs was determinated by MTT assay and the apoptotic of the two drugs was determinated by flow cytometric analysis. Result demonstrated that Paeonia suffruticosa Andr. and Carthamus tinctorius L. induced HUVECs cytotoxity in a concentration-dependent manner. The secondary, oxLDL-induced HUVECs damage was observed by Giemsa stain. When given low concentration of Paeonia suffruticosa Andr. or Carthamus tinctorius L.,the expression of the apoptic body and subG1 induced by oxLDL was reduced. Result demonstrated that Paeonia suffruticosa Andr. and Carthamus tinctorius L.can protect against oxLDL-induced damage.
