Summary:碩士 === 長庚大學 === 生化與生醫工程研究所 === 92 === Hydroxyl-terminated polybutadiene based polyurethane was synthesized by solution polymerization with 1, 4-Butandiol (1, 4-BDO) chain extenders and then modified with N-isopropylacrylamide by UV radiation method (PN). Membranes for wound dressing were prepared by combining chitosan or collagen with modified polyurethane to get chitosan or collagen containing PNCh or PNCo. The effect of N-isopropylacrylamide, chitosan and collagen on the properties of modified polyurethane was studied. Different microstructure of samples could be gotten by processing in 0℃ or -196℃ and modifying with chitosan or collagen. By comparing with PU membranes, the PNCh and PNCo samples contained more saturated water content (1000% ~1500%). The water vapor transmission (WVTR) and permeance for the PNCh and PNCo samples were 900~130 g/day•m2 and 2.4×10-9~4.0×10-9 kg/m2•s•Pa, respectively. The in vitro biodegradation degree of PN membranes increased with increasing the content of N-isopropylacrylamide. The in vitro biodegradation degree of collagen was larger than that of chitosan. From cell culture and cell sheet tests, the membranes prepared in this study were biocompatible and cell can be detached from PN membranes by reducing culture temperature.
