The cardiac cells isolated from 2-day-old SD rat were cultured on the upper level of a millipore transfilter dish with 3.0 μm pores size, and bone marrow stromal stem cells 5-week-old SD rat from were seeded in the lower levels for 7 days. Bone marrow stromal stem cells were observed under phase contrast microscope and stained with five cardiomyocyte markers: α- sarcomeric actin、Troponin T、Nkx2.5、GATA4 and MEF-2 by cardiac cells has no effect on the cell morphology of immunocytochemical technique. Result indicated that bone marrow stromal stem cells. However, after co-cultured with cardiac cells, bone marrow stromal cells expressed cardiac cell marker such as α- sarcomeric actin、Nkx2.5 and GATA4. The factors which induce bone marrow stromal stem cells differentiation to cardiomyocytes could be secreted by cardiac cells and diffuses through millipore transfilter. Cardiac cells may have secreted modulatory factors and induce bone marrow stromal cells to express cardiomyocyte markers, molecular mechanism of this phenomenon deserved further investigation.
