碩士 === 東海大學 === 化學系 === 91 === This thesis reports the use of solid phase peptide synthesis technology for the preparation of three series of novel DNA cleavage and binding agents.Aceording to structure, these pipetides could be distinguished into three category. Series 1 : Peptide-Chlorambucil conjugates. For example : CLB-Asn-Arg-Arg-Trp-Asn-Arg-Arg-Trp-CONH2 (NW8P-CLB) Series 2 : Cyclic peptides. For example : Cyclo[CH2CO-Gly-Tyr-Pro-Arg-Lys-Tyr-Pro-Arg-Lys-Tyr-Pro-Arg-Lys-Cys-CONH2] (CY-14P) Series 3 : Peptides. For example : His-Pro-Arg-Lys-Nmp-Nmp-Nmp-Nmp-Nmp-His-Pro-Arg-Lys-CONH2 (PyH-13P) The synthetic peptides were subjected to agarose electrophoresis to determine its ability to nick supercoiled DNA. The sequence specificity of DNA cleavage or binding by these agents was further studied using high resolution polyacrylamide gel electrophoresis was then employed to study.
